چکیده
Background. Dilated cardiomyopathy (DCM) is a cardiovascular disease of unknown etiology with progressive aggravation. More and more studies have shown that long noncoding RNAs (lncRNAs) play an essential role in dilated formation development. The mechanism action competitive endogenous RNA (ceRNA) networks formed based on the principle lncRNAs affect mRNAs’ expression level by competitively binding microRNAs (miRNAs) has rarely been reported. Objective. This study aimed at constructing lncRNA-miRNA-mRNA ceRNA network bioinformatics analysis methods, discovering, validating potential biomarkers DCM determining possible therapeutic targets from them for drug prediction. Methods. A lncRNA dataset mRNA microarray were downloaded Gene Expression Omnibus Database (GEO). was compared between blood samples patients normal subjects to identify differential mRNAs. constructed using tools, functional pathway enrichment protein-protein interactions performed. mRNAs proteins they encode are then used as predicting drugs. Besides, validated real-time quantitative PCR (qRT-PCR) experiments vitro. Results. differentially expressed successfully established. Two overexpressed key found network: AC093817 AC091062, qRT-PCR further overexpression AC091062. encoded prediction get related Conclusion. supports development cardiomyopathy, AC091062 being cardiomyopathy.

متن کاملBioinformatics identification of miRNA-mRNA regulatory network contributing to lung cancer invasion
Background: Over the past 15 years, significant insights have been gained into the roles of miRNAs in cancer. In various cancers, miRNAs can act as oncogenes, tumor suppressors, or control the metastasis process by modulating the expression of numerous target genes. This study is aimed at determining molecular network of miRNA-mRNA regulating lung cancer invasion, by bioinformatics approaches. ...
متن کاملIdentification of an lncRNA-miRNA-mRNA interaction mechanism in breast cancer based on bioinformatic analysis
Non‑coding RNAs serve important roles in regulating the expression of certain genes and are involved in the principal biological processes of breast cancer. The majority of studies have focused on defining the regulatory functions of long non‑coding RNAs (lncRNAs) and microRNAs (miRNAs/miRs), and few studies have investigated how lncRNAs and miRNAs are transcriptionally regulated. In the presen...
متن کاملConstruction of an oesophageal cancer-specific ceRNA network based on miRNA, lncRNA, and mRNA expression data
AIM To explore the expression profiles of microRNAs (miRNAs), long non-coding RNAs (lncRNAs), and mRNAs in oesophageal squamous cell carcinoma (ESCC) in order to construct an oesophageal cancer-specific competing endogenous RNA (ceRNA) network. METHODS In this work, the expression data of miRNAs, lncRNAs, and mRNAs in ESCC were obtained. An oesophageal cancer-specific ceRNA network was then c...
متن کاملAn Integrated Analysis of miRNA, lncRNA, and mRNA Expression Profiles
Increasing amounts of evidence indicate that noncoding RNAs (ncRNAs) have important roles in various biological processes. Here, miRNA, lncRNA, and mRNA expression profiles were analyzed in human HepG2 and L02 cells using high-throughput technologies. An integrative method was developed to identify possible functional relationships between different RNA molecules. The dominant deregulated miRNA...
